Introduction: Ceritinib is a second-line drug used to treat patients with non-small cell lung cancer (NSCLC) who have acquired resistance to crizotinib. Triazole antifungals are therapeutic choices for patients with cancer to reduce the risk of opportunistic fungal infections. In this study, we investigated whether three CYP3A4 inhibitors of triazole antifungals (voriconazole, itraconazole, and fluconazole) affect the pharmacokinetics of ceritinib in rats. Methods: Ceritinib was orally administered to rats alone or in combination with voriconazole, itraconazole, or fluconazole, and its pharmacokinetic parameters across treatments were compared. The plasma concentration of ceritinib was determined using a validated HPLC-MS/MS method. Results: Co-administration of ceritinib with itraconazole significantly increased the pharmacokinetic parameters (Cmax and AUC) of ceritinib, indicating that itraconazole inhibited ceritinib metabolism and increased systemic exposure. However, no significant effects were observed when voriconazole and fluconazole were used in combination. Conclusions: This study showed that co-administration with itraconazole had a significant effect on the pharmacokinetics of ceritinib, whereas the co-administration of ceritinib with voriconazole or fluconazole had no effect. Dose adjustment of ceritinib should be reconsidered upon co-administration with itraconazole in ongoing clinical practice. Hence, our study provides useful information regarding the combination of ceritinib with the CYP3A4 inhibitors voriconazole, itraconazole, and fluconazole.

Aim This study aimed to demonstrate biosimilarity between Lupin’s Pegfilgrastim versus Neulasta® with respect to pharmacokinetic (PK), pharmacodynamic (PD), immunogenicity, and safety profiles. Methods This double-blind, randomised, single-dose, two-sequence, two-treatment, two-period, crossover, comparative PK and PD study enrolled healthy subjects aged 18 to 45 years. The test product was Lupin’s Pegfilgrastim and the reference product was U.S.-licensed Neulasta®. Blood samples were collected at different time points for assessment of PK, PD, and immunogenicity. Primary objectives included comparing the relative bioavailability and the pharmacodynamic effect of the two products. Secondary objectives included assessing immunogenicity, safety and tolerability. Biosimilarity was concluded if the 90% confidence interval (CI) of the ratio of the geometric least-squares mean (LSM) for PK and PD parameters were within 80 to 125%. Results The geometric mean ratios of Lupin’s Pegfilgrastim and Neulasta® for primary PK parameters were Cmax [94.37 (89.53 to 99.47%)], AUC0-t [94.76 (89.83 to 99.95%)] and AUC0-∞ [94.82 (89.90 to 100.02%)], and for primary PD parameters were ANC_Cmax [100.59 (98.96-102.25%)] and ANC_AUEC0-tlast [100.51 (99.18,101.85%)]. The corresponding 90% CIs were within the pre-defined equivalence limit of 80% and 125%. The incidence of ADA formation was similar between patients receiving Lupin’s Pegfilgrastim and Neulasta®. No new safety concerns were observed. Conclusion This study establishes biosimilarity of Lupin’s Pegfilgrastim and Neulasta® with respect to PK and PD profiles. No clinically meaningful differences were observed between the biosimilar and the reference product in terms of immunogenicity, safety, and tolerability profiles.

Mucormycosis is an infection with vascular inotropism. We report a case of giant cell vasculitis due to a mucormycosis infection. The biopsy of the temporal artery revealed a giant cell vasculitis. A cutaneous biopsy of a necrotic zone confirmed mycormycosis. Despite the treatment, the result was fatal.

Protein allocation determines the activity of cellular pathways and affects growth across all organisms. Therefore, different experimental and machine learning approaches have been developed to quantify and predict protein abundances, respectively. Yet, despite advances in protein quantification, it remains challenging to predict condition-specific allocation of enzymes in metabolic networks. Here we propose a family of constrained-based approaches, termed PARROT, to predict enzyme allocations based on the principle of minimizing the enzyme allocation adjustment using protein-constrained metabolic models. To this end, PARROT variants model the minimization of enzyme reallocation using four different (combinations of) distance functions. We demonstrate that the PARROT variant that minimizes the Manhattan distance of enzyme allocations outperforms existing approaches based on the parsimonious distribution of fluxes or enzymes for both Escherichia coli and Saccharomyces cerevisiae. Further, we show that the combined minimization of flux and enzyme allocation adjustment leads to inconsistent predictions. Together, our findings indicate that minimization of resource, rather than flux, redistribution is a governing principle determining steady-state pathway activity for microorganism grown in suboptimal conditions.Availability and implementation: The implementation of PARROT can be found in the GitHub repository: https://github.com/mauricioamf/PARROT

Research on the variations in soil aggregate stability and ecological stoichiometry at aggregate scales by stand type is of great significance to investigating the distribution, limitation, balance, and cycling of chemical elements. We measured the variations in soil aggregate stability, organic carbon (OC), total nitrogen (TN), and total phosphorus (TP) contents and stocks, in addition to their stoichiometric ratios by stand type (mixed stands of Chinese fir and Mytilaria laosensis and Chinese fir and Michelia macclurei and pure stand of Chinese fir), and aggregate size (<0.25, 0.25-1, 1-2, and >2 mm) in soil profile (0-20 and 20-40 cm) in Guangxi, China. Soil OC and TN contents and C/N, C/P and N/P ratios were more distributed in aggregates of <0.25 mm in the three stands; in contrast, soil OC, TN, and TP stocks were more obvious in aggregates of >2 mm. In comparison to the pure stand, the mixed stands showed significantly higher levels of soil aggregate stability, aggregate-associated OC and TN contents with stocks, TP content, C/N and C/P ratios. The C/N ratio, TN and TP contents, and TP stocks were the key factors in the stand types affecting the stability of soil aggregate changes. Hence, constructing mixed stands and applying appropriate amounts of N and P fertilization can improve soil fertility and promote degraded soil restoration.

Predominantly visual loss, is very rare in Wernicke's encephalopathy. A 22 year old lady, in her 28th week of gestation, presented with a confused mental state, bilateral papilloedema with retinal hemorrhages, ophthalmoparesis, and cerebellar signs. Her MRI brain was suggestive of Wernicke's encephalopathy and she recovered with intravenous thiamine.

Esophageal lung is a rare congenital malformation. We present a full-term infant with an esophageal lung presenting as a white lung, dysmorphic features, and VACTER association. Right pneumonectomy was successfully performed. This is the first report of esophageal lung associated with the VACTER group of anomalies combined with dysmorphic features.

We reported a delayed diagnosised Chinese JATD case with mild skeletal phenotype, and presented with renal insufficiency as the initial symptom of disease onset. The virus SARS-CoV-2 originated in Wuhan, China, in 2019 and has since spread worldwide, infecting humans and animals. Several reports have described the seroprevalence of antibodies in dogs and cats, but there are no reports from Mexico. This study aimed to evaluate the seroprevalence of neutralizing antibodies against the ancestral strain and Omicron BA.1 subvariant in dogs and cats in Mexico. A total of 1111 samples from dogs (n=1052) and cats (n=59) were collected in different regions of Mexico, and the presence of neutralizing antibodies (nAbs) was evaluated by a plaque reduction neutralization test (PRNT90) and microneutralization (MN). The results showed that 15.25% (9 of 59) of cats and 2.85% (30 of 1052) of dogs presented nAbs against the ancestral strain of SARS-CoV-2. However, the analysis of nAbs against Omicron BA.1 in cats showed the same percentage of positive animals but a reduction in the titer. In the case of dogs, 2.09% (22 of 1052) showed nAbs against Omicron BA.1. These results agree with previous reports showing that antibodies are more frequent in cats than in dogs and that these antibodies have a lower capacity to neutralize Omicron BA.1

The comprehensive care that cancer patients require has become increasingly complex and led to the creation of cardio-oncology clinics (COC). Ninety-five patients were referred to our quaternary care COC. Initiation of oral heart failure therapy (OHFT) or advanced cardiac therapies within 1 year following referral were identified and risk factors for these were evaluated. Patients older at the time of cancer diagnosis and with lower LV ejection fraction were more likely to require OHFT. COC’s elicit a high degree of clinically actionable information for pediatric patients suffering from not only cancer, but also cardiomyopathy, a particularly high-risk patient population.

Background: Children with incurable cancer can be treated with metronomic chemotherapy. The aim of this study was to describe the functional status in children with end-stage cancer who received metronomic chemotherapy. Procedure: An observational cohort study was designed with a convenience sample of patients younger than 18 years of age treated with metronomic chemotherapy between 2014-2020 at Hospital Civil de Guadalajara in Mexico. Results: Forty-three patients were included with a median age of 13 years. Metronomic chemotherapy was indicated most frequently in patients with lymphoblastic leukemia in ten cases (23.3%). Before metronomic chemotherapy, more than 70% of patients had disease progression after a second-line treatment. The median treatment duration was 4.2 months. Treatment was suspended in 39 patients, of which 33 patients died. The domain correlated with a lower score on the palliative performance scale at diagnosis was described in 28 cases (65.1%) according to activity level or evidence of disease, in 14 patients (32.5%) with ambulation, and in one patient (2.3%) with intake. According to their score 28/43 (65%) patients presented improvement one month after starting treatment. The McNemar test before and after one month of treatment with metronomic chemotherapy was statistically significant for these symptoms (P=0.00). We found that statistical significance was preserved during the first three months (p = 0.016) of treatment. Conclusions: The outpatient administration, good treatment adherence and few adverse effects make metronomic chemotherapy a valuable treatment option. In our experience, functional status improved significantly in the first three months of treatment.

The sterility of hybrids from interspecific hybridization limits speciation, which widely exists in interspecific hybrids of hermaphroditic plants and also been found in the F1 hybrids of hermaphroditic Argopecten scallops. However, the underlying mechanism of sterility remained unexplored in hermaphroditic animals. In this study, we firstly investigated the mechanism of sterility in the interspecific F1 hybrids of Argopecten scallops by examining the mutations in mitochondrial genes and expressions of nuclear genes. Our results showed that the ATP content in gonads of F1 hybrids was significantly lower than their parents, indicating that energy deficiency may be an immediate cause of sterility in F1 hybrids. The SNP variation types of transition in mitochondrial genes (CYTB, ND4, ND2, ATP6, and COX2) could change their hydrophobicity of amino acids and protein structures, which might contribute to sterility in F1 hybrids. The mutations and rearrangements of mitochondrial genes and abnormal expression of nuclear genes were found in F1 hybrids. Abnormal interaction between the mitochondrial and nuclear genes might contribute to sterility of the F1 hybrids through the following pathways: (1) inhibition of oocyte maturation by enhanced expression of Mfn2; (2) cell cycle arrest in G1 phase of oocytes by inhibited expression of CDK2; (3) cell apoptosis induced by mitochondrial apoptosis; (4) insufficient energy supply from abnormal mitochondria, and (5) mitochondrial dysfunction resulted from abnormal expression of other nuclear genes. The mechanism of sterility in interspecific hybrids of hermaphroditic animals would provide more information for understanding the reproductive isolation and exploitation of heterosis.

Gene transcription and protein synthesis can be widely found in biological research. Based on this viewpoint, this article focuses on a two-dimensional stochastic delay differential equation (SDDE) to investigate the gene expression. A wealth of dynamic behaviors from a mathematical perspective are observed. An optimized method to handle the stochastic delayed system is proposed and we applied it to the model in this paper. Then an averaging Ito equation is obtained. Based on this, we studied the stochastic bifurcation of the system and the corresponding probability distribution with time delay, degradation rate, the intensity of the stochastic process, and the randomization degree as parameters. The numerical results revealed the transition of the probability density function of the system from a Dirac δ function to a nontrivial stationary state. In addition, stochastic P-bifurcation occurs when the presentation of the stationary probability density change from singular peak to crater.

The buffy-tufted-ear marmoset (Callithrix aurita) is a small primate endemic to the Brazilian Atlantic Forest biome, and one of the 25 most endangered primates in the world, due to fragmentation, loss of habitat, and invasion by allochthonous Callithrix species. Using occurrence data for C. aurita from published data papers, we employed model selection and cumulative AICc weight (w+) to evaluate whether fragment size, distance to fragments with allochthonous species, altitude, connectivity, and surrounding matrices influence the occurrence of C. aurita within its distributional range. Distance to fragments with C. jacchus (w+ = 0.94) and non-vegetated areas (w+ = 0.59) correlated negatively with C. aurita occurrence. Conversely, the percentage of agriculture and pasture mosaic (w+ = 0.61) and the percentage of savanna formation (w+ = 0.59) in the surrounding matrix correlated positively with C. aurita occurrence. The findings indicate that C. aurita is isolated in forest fragments surrounded by potentially inhospitable matrices, along with the proximity of a more generalist and invasive species, thereby increasing the possibility of introgressive hybridization. The findings also highlighted the importance of landscape factors and allochthonous congeneric species for C. aurita conservation, besides indicating urgency for allochthonous species management. Finally, the approach used here can be applied to improve conservation studies of other endangered species, such as C. flaviceps, which is also endemic to the Brazilian Atlantic Forest and faces the same challenges.

The kinetics of tautomerization reactions of thiobarbituric acid were thoroughly investigated at the B3LYP/6-311+G(3df,2p) level of theory and elevated temperatures from 270 K up to 1000 K. The kinetic parameters were determined by employing canonical transition state theory (TST). The quantum tunneling effect was included by both wigner and centrifugal-dominant small-curvature semiclassical adiabatic ground-state approximation methods. Intrinsic reaction coordinate (IRC) analysis was performed to certify a unique connection from an obtained transition state configuration to optimized structures of reactant and product. The reaction forces and reaction force constants were computed across the IRC of each reaction. Then, using extremums of reaction forces, each reaction was partitioned into three stages containing reactant, transition state, and product stages. The extent of reactions was followed along with three reaction stages. The dependence of kinetic parameters and curvature of Arrhenius plots on temperature were investigated and discussed in the framework of TST. To further unveil the mechanism of the reactions, the quantum theory of atoms in molecules (QTAIM) was applied. To do this aim, QTAIM descriptors on the BCP of involved bonds including the density of electrons, Laplacian of the density of electrons, Lagrangian kinetic energy, potential energy, and energy density were computed and surveyed along with reaction extent through IRC.
