Understanding how populations diverge and new species arise is a central question in evolutionary biology. ‘Allopatric’ divergence through geographic isolation is considered to be the commonest mechanism generating species biodiversity in mountainous ecosystems. However, the underlying genomic dynamics, especially genomic islands of elevated divergence and genes that are highly diverged as a result of lineage-specific selection, remain poorly understood. Stellera chamaejasme is widely distributed on the Qinghai-Tibet Plateau and in adjacent regions, making it a good model with which to explore genomic divergence in mountainous ecosystems. We assembled a high-quality, chromosome-level genome for this species and re-sequenced the genomes of 24 populations across its distributional range. Our population genomic analyses recovered four distinct genetic lineages corresponding to geographic distributions with contrasting environments. However, we revealed that continuous gene flow occurred during the diversification of these four lineages and inter-lineage hybrids, and plastome introgressions were frequently found in regions of contact. The elevated genomic divergences were highly heterogeneous across the genome. The formation of such genomic islands showed neither correlation with rate of gene flow nor relationship to time of divergence. The lineage-specific positively selected genes potentially involved in local adaptation were found both within and outside genomic islands. Our results suggest that genomic divergence in S. chamaejasme is likely to have been triggered and further maintained by local selection in addition to geographic isolation.

DNA-origami biomanufacturing relies in many cases on the use of asymmetric PCR (aPCR) to generate 500-3500 base, object-specific, single-stranded DNA (ssDNA) scaffolds. Each scaffold is usually purified by agarose gel extraction, a technique that is laborious, limited, not scalable, presents low recovery yields and a low-quality product. Alternatively, we present a chromatography-based method to purify ssDNA scaffolds from aPCR mixtures, which can be used in the context of DNA-origami techniques. aPCR was performed to generate single and double-stranded DNA (dsDNA) from the M13mp18 genome. To isolate the target ssDNA from dsDNA and other PCR impurities, anion-exchange (Q-ligand) and multimodal chromatography (CaptoTM adhere ImpRes) were explored using stepwise gradients with increasing NaCl concentrations. In anion exchange chromatography, the less-charged ssDNA eluted before the dsDNA. In multimodal chromatography, however, the elution pattern was reversed, highlighting the importance played by hydrophobicity. In either case, collected ssDNA-containing fractions were homogeneous and impurity free. Finally, 8.4 μg of a 1000-nt ssDNA fragment were purified and used alongside with site-specific short oligonucleotides (staples) to assemble 63-bp edge length tetrahedrons. Gel electrophoresis showed high assembly yield and purity, whereas fluorescence correlation spectroscopy confirmed that the tetrahedrons had a diffusion coefficient (26.7 μm2 s-1) consistent with the expected size (20 nm).

In this paper, we derive the supercloseness properties and global superconvergence results for the implicit Euler scheme of the transient Navier-Stokes equations. Using a prior estimate of finite element solutions, the properties of the Stokes projection and Stokes operator, the derivative transforming skill and the H-1-norm estimate, we deduce the supercloseness properties of the Stokes projection for the velocity in L∞(H1)-norm and pressure in L∞(L2)-norm. Then the supercloseness properties of the interpolation operators are obtained for two pairs of retangular element: the bilinear-constant element and the Bernadi-Raugel element. Finally, by the interpolation postprocessing technique, we obtain the global superconvergent results. The supercloseness analysis is based on the Stokes projection, which makes the proof more concise. Compared with previous superconvergence results, a lower regularity of solution is needed, and no time step restrict is required.

The rapid and successful strides in quantum chemistry in the past decades can be largely credited to a conspicuous synergy between theoretical and computational advancements. However, the architectural computer archetype that enabled such a progress is approaching a state of more stagnant development. One of the most promising technological avenues for the continuing progress of quantum chemistry is the emerging quantum computing paradigm. This revolutionary proposal comes with several challenges, which span a wide array of disciplines. In chemistry, it implies, among other things, a need to reformulate some of its long established cornerstones in order to adjust to the operational demands and constraints of quantum computers. Due to its relatively recent emergence, much of quantum computing may still seem fairly nebulous and largely unknown to most chemists. It is in this context that here we review and illustrate the basic aspects of quantum information and their relation to quantum computing insofar as enabling simulations of quantum chemistry. We consider some of the most relevant developments in light of these aspects and discuss the current landscape when of relevance to quantum chemical simulations in quantum computers.

The hypersingular integral equation has been studied widely in boundary element methods, especially in natural boundary element methods. The asymptotic expansion of error function of composite rectangle rule for the computation of Hadamard finite-part integrals with the hypersingular kernel $ 1/\sin^{2}(x-s)$ is obtained. Extrapolation algorithm is constructed. In order to solve the hypersingular integral equation, the superconvergence point is taken as the collocation point, then the extrapolation algorithm is presented and the convergence rate of extrapolation algorithm for hypersingular integral equation is presented. At last, some numerical results are also illustrated to confirm the theoretical results and show the efficiency of the algorithms.

An improved swing-enhanced floating-inverter amplifier (SEFIA) using the correlated-level-shifting (CLS) technique is presented, simultaneously achieving both high gain and high output swing. The proposed SEFIA uses a reused capacitor to replace the dynamic biasing capacitor and input coupling capacitor. One of the two reservoir capacitors, which supplies power to the amplifier in the auto-zero phase, is also reused in the level shifting phase. Post-layout simulation results show that in the 180 nm CMOS process; the DC gain remains above 73dB for clock frequencies between 50 kHz to 400 kHz. Compared to conventional FIA with CLS, the proposed SEFIA with a 1.8V supply achieves the 1.54 times output swing increase at a clock frequency of 250 kHz. The CLS technique boosts the DC gain of the proposed SEFIA from 50dB to 74.6dB.

African swine fever virus (ASFV) causes contagious hemorrhagic disease of pigs with high morbidity and mortality. To identify the molecular characterization of ASFV strains circulating in Guangxi province, southern China, tissue samples collected from domestic pigs that died as a result of severe hemorrhagic disease during 2019-2020 were detected for ASFV, and 66 representative ASFV strains from Guangxi province were genetically characterized by sequence analysis of the C-terminal region of B646L (p72) gene, the complete E183L (p54) gene, the variable region of EP402R (CD2v) gene, the central variable region (CVR) of B602L gene, and the tandem repeat sequence (TRS) within the intergenic region (IGR) between the I73R and I329L (I73R/I329L) genes. Phylogenetic analysis revealed that the ASFV strains from Guangxi province belonged to genotype I and II based on the p72 and p54 genes, and there were eight different tetrameric TRS variants based on the CVR of B602L gene. Phylogenetic analysis of the CD2v gene revealed that these ASFV strains belonged to serogroup IV and VIII. Eight of the 66 strains belonged to genotype I and serotype VIII, and showed deletion of whole MGF505-2R gene. The sequence analysis of the IGR between the I73R/I329L genes showed that IGR II and III variants were co-circulating in Guangxi province. The results indicated that ASFV strains circulating in Guangxi province during 2019-2020 outbreaks showed highly genetic diversity, of which genotype I and II strains, as well as serogroup IV and VIII strains, were simultaneously epidemic, and there existed wild-type strains and naturally gene-deleted strains in the field. This is the first report on the molecular characterization of the ASFV strains circulating in southern China.

Background and Purpose: CKD is a clinical syndrome with slow development and gradual deterioration of renal function. At present, modern medicine still lacks an ideal treatment method for this disease, while TCM has accumulated rich clinical experience in the treatment of CKD, which can effectively improve renal function and delay renal failure, and has unique advantages. RC is widely used in clinical practice to treat CKD, especially the ‘Kidney-Yin’ deficiency syndrome. However, the compatibility mechanisms responsible for its effects in experimental studies, including preclinical and clinical researches, are still not fully understood. Experimental Approach: Adenine-induced CKD rats were used to investigated the preventive effect of RC on CKD rats. According to the Illumina-based high-throughput 16S rRNA gene sequencing results, to discuss the flora abundance of rats in different treatment group. Using Chamber, Western blot and PCR were used to investigate the effects of RC on intestinal barrier function and its molecular mechanism in model animals. H&E and M&T staining diagram were used to observe the pathological state of renal tissue in different group. ELISA was used to study the expression of inflammatory factors in different treatment group. Conclusion and Implications: We obtained evidence showing an obvious effect of RC on strengthening the intestinal barrier and modulating gut microbiota in adenine-induced CKD rats. This project revealed the compatibility mechanism of RC in regulating intestinal microecology and barrier function to intervene in CKD, and provided basis and ideas for the clinical application of RC and the development of innovative drugs for CKD.

Aim: High-dose methotrexate (HD-MTX) therapy is commonly used in acute lymphoblastic leukemia (ALL) which need drug monitoring. This study was to analyze the influence factors and prognostic role of the plasma MTX concentration in ALL. Methods: 1435 HD-MTX courses of 246 childhood ALL were enrolled. MTX doses were 3 g/m2 for low-risk (LR) group and 5 g/m2 for intermediate or high-risk groups. The target 24-hours (24h) MTX concentrations were set at 33 μmol/L for LR and 65 μmol/L for non LR group. Results: The median 24h MTX concentrations were 42.0 μmol/L for LR and 70.0 μmol/L for non LR group. Only SLCO1B1 genotype in the LR group and age in the non LR group was associated with the 24h MTX concentrations. The survival results were comparable between patients with or without courses failed to reach the target 24h MTX concentration. MTX excretion delay was observed in 211/1435 (14.7%) courses of 125 patients, which more commonly caused MTX-induced toxicities. All the 6 CNS relapses occurred in patients with MTX excretion delay, while in those without did not have any type of CNS relapse (6/119 vs. 0/121, P=0.015). Patients with more than 20% of MTX excretion delay courses had significant worse survival (83.8%±5.0% vs. 93.7%±1.8% for EFS, P=0.014, and 88.8%±4.3% vs. 97.2%±1.1% for OS, P=0.010). Conclusions: Achieving the target 24h MTX concentration during HD-MTX therapy is not the useful indicator to predict the survival of ALL patients. MTX excretion delay is an independent factor for predicting the CNS relapses and worse prognosis.

Porcine circovirus type 3 (PCV3) is an emerging pathogen, that has been reported worldwide in all ages of healthy and clinically ill pigs. The presence of this virus in Hungary has been confirmed in a commercial farm experiencing reproductive failures, but there were no data on the circulation of PCV3 in the country. Here we report the prevalence and the genetic diversity of PCV3 in Hungarian herds. For the estimation of the prevalence altogether 1855 serum samples, 176 oral fluid and 97 processing fluid samples were collected in a systematic, cross-sectional method from 20 large scale swine herds, and tested by real-time qPCR. PCV3 was present in at least one type of diagnostic matrix in 19 out of the 20 (95%) pig farms. The highest detection rates were observed in the processing fluid samples (61%), but 41% of the oral fluid and 23% of the serum samples were also positive. The virus was found in all age groups and slightly more adult animals were infected than growing pigs, but the viral burden was lower amongst them. Phylogenetic analysis of nine full genomes, obtained from either the sampled herds or organ samples of PCV3 positive carcasses showed high nucleotide identity between the detected sequences, which all belonged to PCV3a genotype. Our results indicate that PCV3 is widespread in Hungary but in most cases the virus seems to circulate subclinically, infecting all age groups and production phases without the presence of apparent clinical disease.

Individuals must be aware of different opportunities and possibilities in order to grow as individuals, and diversity allows this to happen. As diversity prepares individuals for a diverse world, it is also important to students' learning. Ultimately, diversity contributes significantly to the creation of new knowledge due to the fact that it helps people draw on their own experiences and the experiences of others in order to discover new approaches to problems.

Objective: To critically evaluate the Quality Management System (QMS) for Clinical Nutrition (CN) in Jiangsu. Monitor its performance in quality assessment as well as human resource management from nutrition aspect. Investigate the appliance and development of Artificial Intelligence (AI) in medical quality control. Data Source: The study source of this research was all the staffs of 70 Clinical Nutrition Department (CND) of the tertiary hospitals in Jiangsu Province, China. These departments are all members of the Quality Management System of Clinical Nutrition in Jiangsu (QMSNJ). Methods: An online survey was conducted on all 341 employees within all these CNDs based on the staff information from the surveyed medical institutions. The questionnaire contains 5 aspects, while data analysis and AI evaluation were focused on human resource information. Results: 330 questionnaires were collected with the respondent rate of 96.77%. The QMS for CN has been build up for CNDs in Jiangsu, which achieved its target in human resource improvements, especially among dietitians. The increasing number of participated departments (42.8%) and the significant growth of dietitians (p=0.02, t=-0.42) are all expressions of the advancements of QMSNJ. Conclusion: As the first innovation of an online platform for QM in Jiangsu, JPCNMP has been successfully implemented among QMS from this research. This multidimensional electronic system can help QMSNJ and CND achieve quality assessment from various aspects, so as to realize the continuous improvement of clinical nutrition. The instrument of online platform, as well as AI technology for quality assessment is worth to be recommended and promoted in the future.

Background:Juvenile myelomonocytic leukemia (JMML) is a rare hematological malignancy in young children and can only be cured through the allogeneic stem cell transplantation. Procedure:We have retrospectively analyzed the outcomes of nine children with JMML after unrelated cord blood transplantation (UCBT). Results:Eight patients who have received a myeloablative conditioning regimen of fludarabine (FLU), busulfan (BU), and cyclophosphamide (CY) have gotten engraftment. None of the nine patients has relapsed following initial UCBT. Six patients are still alive and in complete remission after UCBT with a median observation time of 43 months (range: 10–80 months). The 5-year overall (OS) and event-free (EFS) survival rates are 77.8% and 66.7%, respectively. Conclusions:This study shows that UCBT with FLU–BU–CY conditioning regimen can represent a suitable option for children with JMML.

Transcatheter aortic valve replacement (TAVR) is a revolutionized treatment for severe aortic valve stenosis. Although new and improved TAVR devices are constantly being developed, cardiac conduction abnormalities post-TAVR requiring permanent pacemaker implantation (PPMI) still occur frequently. Previously, pre-existing right bundle branch block (RBBB) has been shown to be predictive of PPMI after TAVR, while occurrence of new left bundle branch block (LBBB) was associated with a higher rate of PPMI. However, less attention has been paid to the clinical values of new-onset RBBB post-TAVR. To our knowledge, this is the first report focus on the association of new-onset RBBB and PPMI after TAVR. Sometimes only changes in the right bundle branch can be detected, but the patient may have co-existing severe conduction block at this time, which caused adverse events including Adams-Stokes syndrome. New-onset RBBB post-TAVR may also have important clinical implications.

Background: In the face of this threatening epidemic, the health behaviors of people gain great importance in terms of both minimizing the prevalence and reducing possible life losses. perception of health affects health behavior. Psychological resilience is key in combating a challenging situation that threatens both physical and psychological health. Objective: This study was aimed to examine the correlation between psychological resilience and health perception in terms of some variables in the case of the COVID-19 pandemic. Methods: This cross-sectional study was carried out using quantitative and general screening models. In data collection, an Individual Information Form, the Brief Resilience Scale (BRS) and the Perception of Health Scale (PHS) were used. Results: The participants had a mean age of 33.24 ± 10.95, with 35.7% males. Mean scores were 52.68 ± 6.80 for PHS and 20.11 ± 4.24 for BRS. We found that the health perceptions of the participants increased in parallel with their psychological resilience (p<0.05). Conclusion: The participants were found to have moderate levels of health perception and psychological resilience. Their measures against the epidemic and certain socio-demographic characteristics led to differences in their psychological resilience and health perceptions. Knowing the psychological resilience and health perceptions of individuals can improve the quality of treatment and care.

Objective: To compare postoperative reproductive outcomes between salpingectomy and neosalpingostomy for bilateral severe hydrosalpinges. Design: Single center, prospective cohort study. Setting: A hospital affiliated to a medical college in China. Population: Women aged 20 to 45 years old, diagnosed with bilateral hydrosalpinges and scheduled for surgery were initial eligibility criteria. Women with previous tubal surgery or tubal pregnancy, no fertility intention, or no confirmation of severe bilateral hydrosalpinges during surgery were excluded. Methods: Bilateral salpingectomy or neosalpingostomy was performed based on a shared decision approach. Main outcome measures: The primary outcome was the cumulative livebirth rate. Secondary outcomes included time to first live birth, biochemical pregnancy, clinical miscarriage, ectopic pregnancy, mode of conception, and gestational age at delivery. Results: A total of 113 women were involved in the analysis. When the result of In Vitro Fertilization (IVF) in the neosalpingostomy group was incorporated, salpingectomy resulted in a higher cumulative livebirth rate (85.3% vs 76.0%, hazard ratio of the whole survival curve = 2.18, 95% CI 1.37 - 3.45), a lower risk of ectopic pregnancy (1.8% vs 20.7%, risk ratio = 0.07, 95% CI 0.01 - 0.57), and a shorter time to live birth than neosalpingostomy. 16/58 (27.6%) women in the neosalpingostomy group had a live birth via natural conception, compared to 0/55 (0.0%) in the salpingectomy group. Conclusions:Salpingectomy for bilateral severe hydrosalpinges resulted in a higher cumulative livebirth rate and a lower risk of ectopic pregnancy. However, neosalpingostomy can offer certain option for women to conceive naturally without IVF treatment.

Feathers account for the major pollutant of the poultry industry due to its recalcitrant nature. They are composed of keratins that has numerous application in various fields. The untreated feathers production from poultry industry are considered major pollutant because of their resistance towards protease degradation. Chicken feathers can be enzymatically degraded by keratinases that are secreted by numerous bacteria and fungi and converted into value-added products. Degradation by keratinolytic bacteria represent as an alternative for the development of cost-effective, eco-friendly and cheap source of nitrogenous fertilizers that are needed for plant growth. The feather degradation also results in the release of tryptophan that acts as precursor for the phytohormone. The hydrolysate plays important role in the seed germination and plant growth. They also serve as an alternative tool to improve soil biological activities, organic farming and agro-ecosystems. The mechanism of degradation comprises of the three steps, sulfitolysis, proteolysis that is followed by deamination.
