We have reported a gal mutant called galE stop0, wherein the galE stop codon was changed to a sense codon. The experiment results demonstrated that preventing galE translation termination inhibited the production of galE 3’ ends. This implies that when the galE translation termination was prevented, the galE 3’ ends generation was reduced or impaired. We anticipated that the translation of galE would continue to galT, producing a chimeric protein GalE-GalT. This study verified that the chimeric protein was produced, but unexpectedly, we found that the GalT protein was also synthesized in the mutant, and its amount equaled that in the wild-type. In the wild-type, we also found that the GalE-GalT chimeric protein was produced in an amount equal to that of the GalE protein. These results suggest that translation termination of galE and translation initiation of galT occur independently, thus, corroborating our transcription-translation model: At the cistron junction, transcription, decoupled from translation due to the translation termination of galE, needs translation initiation of galT to continue downstream; otherwise, transcription would be terminated by Rho.

Background and Aims: The extravascular implantable cardioverter defibrillator (EV ICD) has an extended projected battery longevity compared to the subcutaneous implantable cardioverter defibrillator (S-ICD). This study used modeling to characterize the need for generator changes, long-term complications, and overall costs for both the EV ICD and S-ICD in healthcare systems of various countries . Methods: Battery longevity data were modelled using a Markov model from averages reported in device labelling for the S-ICD and with engineering estimates based on real life usage from EV ICD Pivotal Study patient data to introduce variability. Clinical demographic data of recipients were derived from published literature. The primary outcomes were defined as the number of generator replacement surgeries, complications, and total healthcare system costs due to battery depletion over the expected lifetime of patients receiving EV ICD or S-ICD therapy. A one-way sensitivity analysis of the model was performed for the US healthcare system. Results: Average modelled battery longevity was determined to be 7.3 years for the S-ICD compared to 11.8 years for the EV ICD. The probability of a complication after a replacement procedure was 1.4%, with an operative mortality rate of 0.02%. The use of an EV ICD was associated with 1.4-1.6 fewer replacements on average over an expected patient lifetime as compared to an S-ICD and a 24.3-26.0% reduction in cost. The US sensitivity analysis found use of an EV ICD resulted in a reduction in replacement surgeries of greater than 1 (1.1-1.6) along with 5-figure cost savings in all scenarios ($18,602-$40,948). Conclusion: The longer projected battery life of the EV ICD compared to the S-ICD has the potential to meaningfully reduce long-term morbidity and healthcare resources related to generator changes from the perspective of multiple diverse healthcare systems.

Molecular assays from nasopharyngeal swabs are the current reference method to diagnose COVID-19. As an alternative, we evaluated the performance of the iFlash-2019-nCoV Antigen® (YHLO, Shenzhen, China), developed for SARS-CoV-2 N-antigen detection in serum samples. Specificity, determined on 50 pre-pandemic samples, was 100%. Overall sensitivity, evaluated on 40 sera from patients with RT-PCR confirmed infection, was 67.5%. However, sensitivity reached 73% in symptomatic patients, 80% in patients with high and medium nasopharyngeal (NP) viral loads (samples with Ct≤33) and, 90% in samples collected within the first week after symptoms onset. These sera were further analyzed with the COV-QUANTO® ELISA and COVID-VIRO® LFIA assays (AAZ, Boulogne-Billancourt, France). EIA Ag assays from Yhlo and AAZ had comparable performances, and both were more sensitive than the LFIA. These findings suggest that SARS-CoV-2 N-antigen detection in serum could be an alternative to PCR from NP swabs, at least early after onset of symptoms. Further studies are required to confirm these results.

Background: Anaplastic large cell lymphoma (ALCL) is an uncommon form of Non-Hodgkin Lymphoma (NHL) in younger patients, accounting for less than 15% of cases. The median age at presentation is 12 years and is exceedingly rare in infants. Various prognostic factors, risk stratification and novel treatment strategies have been studied in pediatric ALCL. There is no separate data available on the disease biology, treatment regimens and prognostic factors among infantile ALCL. Outcomes for infants with ALCL have been reported to be inferior to that of older children. The long-term Event Free Survival (EFS) in general among pediatric patients is approximately 70% regardless of the treatment approach. We report a case of an 8month old infant diagnosed with ALK positive ALCL with visceral organ involvement stratified as high risk and treated according to ALCL 99 protocol with excellent clinical response, on regular follow up and is disease free since 2years.

Three-dimensional (3D) cell cultures are to date the golden standard in biomedical research fields due to their enhanced biological functions as compared to conventional two-dimensional (2D) cultures. 3D cell spheroids as well as organoids are better suited to replicate tissue functions, which enables their use both as in vitro models for basic research and toxicology, as well as building blocks used in tissue/organ biofabrication approaches. Culturing 3D spheroids from bone-derived cells is an emerging technology for both disease modelling and drug screening applications. Bone tissue models are mainly limited by the implementation of sophisticated devices and procedures that can foster a tissue-specific 3D cell microenvironment along with a dynamic cultivation regime. In this study, we consequently developed, optimized and characterized an advanced perfused microfluidic platform to improve the reliability of 3D bone cell cultivation and to enhance aspects of bone tissue maturation in vitro. Moreover, fluid shear stress, generated by fluid flow inside the chamber, was used to mimic the dynamic cell environment in bone architecture and thus improve an osteogenic 3D microenvironment in the multifunctional spheroid-array platform. The 3D cell cultures cultivated in our bone on a chip model exhibited increased mineralization and viability compare to static conditions. Thus, this system represents a powerful tool how to study bone physiology and pathophysiology in vitro.

Dextrocardia is a cardiac positional anomaly in which the heart is located in right hemithorax with base-to-apex axis directed to the right and caudad. A number of congenital heart defects have been reported with dextrocardia, including VSD, PDA, ASD, TOF, pentalogy of Fallot, infundubular PS, transposition and pseudotruncus and total anomalous pulmonary venous return. We will share a patient with severe TR due to tricuspid valve commissural prolapse. A 42-year-old female patient was admitted to our clinic with the complaint of dyspnea. Transthoracic echocardiography revealed dextrocardia and severe TR consisting of two separate regurgitation jets. Contrast echocardiography performed due to dilatation of the coronary sinüs, did not show persistent left superior vena cava, and no right-to-left shunt was observed. Transesophageal echocardiography showed a prolapse in the commissure where the tricuspid anterior and septal leaflets meet, and a severe eccentric regurgitation jet with an area of vena contracta 0.75cm2 in the 3D MPR was observed. A moderate regurgitation jet was also seen from the coaptation line of all three leaflets. Commussural prolapse and regurgitation jet revealed in detail by 3D imaging. No significant pathology was detected in the other valves except mild insufficiency. Right heart catheterization and tricuspid valve surgery were planned for the patient with normal right heart functions. Although it is known that there are many congenital pathologies accompanying dextrocardia, we are happy to share our experience with you as the first case to report the coexistence of primary tricuspid valve disease and commusural prolapse with 3D detailed imaging.

Suboptimal response to cardiac resynchronization therapy (CRT) may be improved by optimization of device parameters using echocardiography. For this purpose, the aortic velocity-time integral (aVTI) was used as a target metric to define optimal velocity timings for each ventricle. dP/dt is a parameter used for the assessment of myocardial contractility. In this study, we aimed to evaluate the effectiveness of Doppler-derived dP/dt in optimization by assessing the possible correlation between aVTI and dP/dt. Patients with CRT admitted for routine follow-up were included in the study. Aortic VTI and dP/dt measurements were recorded in four different standard pacing configurations during reprogramming. A total of 46 patients were included in the final analysis. No correlation was found between the aVTI and the delta dP/dt value in the two configurations where the change in dP/dt was maximum (p=0.894). In the two configurations where the change in aVTI was maximum, there was also no correlation between the delta dP/dt and the delta aVTI (p=0.715). When patients were dichotomized according to the median value of dP/dt, there were no differences in aVTI, NYHA classes, LVEF, and mitral regurgitation (MR) severity (p=0.4; p=0.5; p=0.7; p=0.3; respectively). The change in both dP/dt and aVTI was statistically significant when switching from RV-only to QRS width-targeted configuration (p=0.001; p=0.041; respectively). In conclusion, aVTI recorded at different pacing configurations was not correlated with dP/dt during interventricular optimization. However, both parameters consistently showed a positive effect of biventricular pacing on contractile synchronization and stroke volume.

The 2030 Agenda offers a list of global environmental, social, and economic objectives to attain sustainable development. However, achieving the Sustainable Development Goals (SDGs) is challenging given the complex interactions between different SDGs and their spillover effects. System dynamics models have the capacity to integrate multisectoral dynamics of SDG interactions. We developed a system dynamics model-the Local Environmental and Socio-Economic Model (LESEM)-to analyse and quantify context-based SDG interactions at the local scale using a participatory model co-design process with local stakeholders. The LESEM was developed for a case study in the Goulburn-Murray Irrigation District in northern Victoria, Australia. We present an illustrative application of the model that quantifies SDG interactions across four high-priority SDGs, namely clean water and sanitation (SDG 6), agricultural activities (SDG 2), economic growth (SDG 8), and life on land (SDG 15). Our results suggest that agricultural land area may shrink by 62,522 ha due to the decline in water resource availability (SDG 6) under a business-as-usual (BAU) scenario from 2022 to 2050. However, the results also highlight that agri-food production (SDG 2) is likely to increase due to intensification to meet future agri-food demand, and higher values of farm output may improve local prosperity. The projections also suggest that environmental pressures may increase due to increasing agricultural intensification and reduced water availability. The LESEM facilitates integrated and strategic decision-making and helps local policymakers identify and quantify potential trade-offs and synergies that benefit multiple SDGs, which eventually leads local communities toward sustainability.

Malignant rhabdoid tumors (MRT) are very aggressive neoplasms seen in infantile age groups. A high index of suspicion is needed for early diagnosis. They are small round cell tumors with characteristic loss of INI -1 expression. Here we present a case series of 6 rhabdoid tumors diagnosed over 2 years, of which 4 of them presented as soft tissue swelling neck and chest wall. All of them had a very short duration of presentation and despite timely diagnosis, 4 of them progressed while on treatment and resulted in mortality.

INTRODUCTIONAs a subtype of biliary tumors, gallbladder cancer is rare yet highly malignant1. Characterized by its ease of metastasis and poor prognosis, its common sites of metastasis include lymph nodes and the liver. However, it can also spread to other organs and tissues such as the lungs, bones, and peritoneum2. It’s critical to note that the incidence of lung metastasis in gallbladder carcinoma hovers around 10-30%, with a dismal 5-year survival rate of around 5-10%3-4. Unusually, when gallbladder carcinoma metastasizes to the lungs, it can sometimes present as pulmonary cavitary lesions, although this manifestation is not commonly observed. In this light, we present a case of gallbladder carcinoma with pulmonary manifestations of cavitary lesions.

Engineering high-performance and low-cost bifunctional catalysts for H2 (HER) and O2 (OER) evolution under industrial electrocatalytic conditions remains challenging. Here, we use the stronger electronegativity of rare-earth yttrium ion (Y3+) to in-situ induce NiCo layered double hydroxide (LDH) nanosheets from the NiCo foam (NCF) treated by a dielectric barrier discharge plasma (PNCF), and then obtain a nitrogen-doped YNiCo phosphide (N-YNiCoP/PNCF) after the phosphating process using RF plasma in nitrogen. The obtained N-YNiCoP/PNCF has a large specific surface area, rich heterointerfaces and the optimized electronic structure, causing a high electrocatalytic activity in HER (331 mV Vs 2000 mA cm-2) and OER (464 mV Vs 2000 mA cm-2) reactions in 1M KOH electrolyte. The X-ray absorption spectroscopy (XAS) and DFT quantum chemistry calculations reveal that the coordination number of CoNi decreased with the incorporation of Y atoms, which cause a much shorter bonds of Ni and Co ions and achieving a long-term stability of N-YNiCoP in HER and OER under the simulated industrial conditions. Meanwhile, the CoN-YP5 heterointerface formed by the plasma N-doping is the active center for overall water splitting. This work opens a new direction for designing high-performance rare-earth elements into engineering bifunctional electrocatalysts in the renewable energy field.

Metal ion doping is the most viable method to modulate the optical properties of lead-free Cs2AgInCl6 double perovskite (DP) nanocrystals (NCs). However, despite multiple cations doping can allow for emission color tuning, their random distribution into the perovskite matrix poses severe limits caused by uncontrollable and adverse energy interactions between the dopants, resulting in poor emission color purity and photoluminescence (PL) quenching. To address this issue, we designed Cs2AgInCl6-based core-shell NCs doped with different metal ions in the two layers via an epitaxial growth strategy, with the aim to spatially separate the diverse optically-active centers. We selected Cs2AgInCl6:Bi as the core and Cs2AgInCl6:Yb, Er as the shell, and successfully constructed Cs2AgInCl6:Bi@Cs2AgInCl6:Yb, Er core-shell NCs. The so-designed core-shell NCs allow for non-interacting dual-modal luminescence (upconversion/downshifting) yielding not only broadband self-trapping excitons orange emission under ultraviolet (UV) excitation, but also upconversion green light emission under near-infrared (NIR) excitation, so far unprecedented in DP NCs. Taking advantage of the high photostability and stability against moisture bestowed by the shell coating, we demonstrate that the resulting core-shell DP NCs can be successfully employed as luminescent ink for dual-mode anti-counterfeiting labels. These results not only provide a novel design strategy for multi-emissive lead-free NCs, but can also represent a milestone to realize controllable luminescent materials for anti-counterfeiting and information storage.

Vagally-mediated heart rate variability (vmHRV) at resting-state has been associated to cognitive functions dependent on cognitive control, such as memory. However, little is known about the phasic interaction between cognitive and autonomic control. In a pre-registered within-between-subject designed experiment, the potential of vmHRV biofeedback to simultaneously stimulate vmHRV during memory processing and cognitive control over memory was tested, along with investigating psychophysiological association. 71 young healthy adults completed (twice) a false memory task in virtual reality. Immediately before memory encoding and retrieval, participants practiced either vmHRV biofeedback or a control breathing exercise. Cognitive control over memory was assessed as the confidence towards false memories and the capability to discriminate them from true memories. Resting-state vmHRV before each test and phasic vmHRV during memory encoding and retrieval was measured as the root mean square differences (RMSSD) in heart period. vmHRV biofeedback had neither an immediate effect on cognitive control over memory nor on phasic RMSSD. Both metrics were associated only under consideration of the resting-state and heart rate values. Cognitive control over memory was positively predicted by parallel reactivity (i.e., change from baseline) in heart rate (ß = .333) and RMSSD (ß = .238) at memory retrieval. In consistence with previous psychophysiological models, the findings demonstrate a link between cognitive and phasic vagally-mediated autonomic control which extends to higher-level cognitive functions such as long-term memory. In this context, memory performance seems to be dependent on tonic and phasic (frequency) components of parasympathetic modulation in response to memory processing.

Convalescent plasma samples that can be collected from individuals after the resolution of infection and vaccination are an invaluable source of neutralization antibodies against the virus. Although plaque reduction assay with replicative virus is the gold standard of analyzing neutralization potency of convalescent plasma, it is a technically demanding procedure requiring high biosafety level (BSL-3/4) laboratory and equipment. The abundance of neutralizing antibodies varies among individuals, therefore fast and reliable methods to identify neutralization potency of plasma samples are needed. In this paper, G-protein deficient vesicular stomatitis virus (VSV-ΔG) carrying a C-terminal 21 amino acid truncated severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) Spike protein was generated for pseudovirus neutralization assay. We analyzed SARS-CoV-2 neutralizing potency of vaccinated human convalescent plasma samples (n=13) and plasma samples of healthy people (n=2). Human convalescent plasma samples were examined against the ancestral Wuhan strain and two SARS-CoV-2 variants (B.1.1.7, and B.1.351) using a VSV-ΔG-Sdel21 pseudovirus and Vero E6 cell line. Neutralization values against pseudotyped virus were compared to those of plaque reduction assay against authentic virus. The serum neutralizing titer of convalescent plasma measured by pseudovirus assay has a good correlation with that measured by plaque reduction assay (R 2= 0.7). The pseudovirus assay is safer and timesaving than the replicative virus-based plaque reduction assay, and has several advantages in evaluating a new vaccine, newly emergent variants, and approved vaccine efficacy against variants of concern as well as in viral fusion-focused treatment analysis that can be performed under BSL-2 conditions.

Macroscopic chiral spherulites prepared by hierarchical self-assembly have attracted considerable attention due to their excellent property as chiroptical materials. However, preparing controllable handedness spherulites in bulk film remains a challenge due to the absent knowledge of the evolution mechanism from the molecule to macroscopic crystal during chiral assembly. In this contribution, we constructed chiral controllable macroscopic spherulites with circularly polarized luminescence (CPL) using enantiomeric tartaric acid and Rhodamine B co-assembled with liquid crystal block copolymers, poly (ethylene oxide)-b-poly (methyl methacrylate) bearing azobenzene group side chains. It was found that the chiral liquid crystal field induced by exogenous chiral molecules was closely related to the formation of macroscopic chiral spherulites. Moreover, the transformation of azobenzene cis-trans isomerization under photo-thermal endows films with adjustable CPL. This facile strategy provides a platform to design large-scale chiral structures for chiroptical switching, encryption, and memory storage materials.

Conservation of breeding seabirds typically requires detailed data on where they feed at sea. Ecological niche models (ENMs) can fill data gaps, but rarely perform well when transferred to new regions. Alternatively, the foraging radius approach simply encircles the sea surrounding a breeding seabird colony (a foraging circle), but overestimates foraging habitat. Here, we investigate whether ENMs can transfer (predict) foraging niches of breeding tropical seabirds between global colonies, and whether ENMs can refine foraging circles. We collate a large global dataset of tropical seabird tracks (12000 trips, 16 species, 60 colonies) to build a comprehensive summary of tropical seabird foraging ranges and to train ENMs. We interrogate ENM transferability and assess the confidence with which unsuitable habitat predicted by ENMs can be excluded from within foraging circles. We apply this refinement framework to the Great Barrier Reef (GBR), Australia to identify a network of candidate marine protected areas (MPAs) for seabirds. We found little ability to generalise and transfer breeding tropical seabird foraging niches across all colonies for any species (mean AUC: 0.56, range 0.4-0.82). Low global transferability was partially explained by colony clusters that predicted well internally but other colony clusters poorly. After refinement with ENMs, foraging circles still contained 89% of known foraging areas from tracking data, providing confidence that important foraging habitat was not erroneously excluded by greater refinement from high transferability ENMs nor minor refinement from low transferability ENMs. Foraging radii estimated the total foraging area of the GBR breeding seabird community as 2,941,000 km2, which was refined by excluding between 197,000 km2 and 1,826,000 km2 of unsuitable foraging habitat. ENMs trained on local GBR tracking achieved superior refinement over globally trained models, demonstrating the value of local tracking. Our framework demonstrates an effective method to delineate candidate MPAs for breeding seabirds in data-poor regions.

In this paper, we consider the reflected backward stochastic differential equations driven by G-Brownian motion with two reflecting obstacles, whose generators satisfy a kind of non-lipschitz condition in y. The existence and uniqueness can be obtained by the method of Picard iteration, a penalization method and some uniform estimates.
